Atomic view of a histone chaperone

ScienceDaily (Mar. 2, 2012) ? Mayo Clinic researchers have gained insights into the function of a member of a family of specialized proteins called histone chaperones. Using nuclear magnetic resonance (NMR) spectroscopy and X-ray crystallography, they have determined the 3-D structure and interactions of the histone chaperone Rtt106 down to the atomic details.

The findings are published in the journal Nature.

"The interactions we described are important for gene silencing and the DNA damage response," says senior author Georges Mer, Ph.D., a Mayo Clinic structural biologist. "This is exciting because our newfound knowledge will help us better understand these fundamental cellular processes."

In cells, our DNA is part of a structure called chromatin, composed of proteins, the majority called histones, which are wrapped with the DNA. Associated with the histones is another group of proteins called histone chaperones, which promote the proper assembly or disassembly of the chromatin during the times our DNA is replicated or repaired when damaged. Their dysfunction has been linked to cancer, aging and other diseases.

The Discovery

Before this Mayo study, scientists knew that the histone chaperone Rtt106 helped in the deposition of histones -- specifically, a complex of histones H3 and H4 -- onto the replicating DNA. They did not understand how Rtt106 does this, given that it does not possess any of the known requirements. Histone H3 is in a modified form where one of its amino acids, lysine 56, is acetylated. Rtt106 does not seem to have an acetylated lysine reader domain.

Mayo researchers discovered two novel domains in Rtt106 that take on this role. One, termed the homodimerization domain, allows two molecules of Rtt106 to be linked so they can cooperate in binding histones H3 and H4. The other, called the double PH domain, is responsible for sensing the acetylated lysine of H3 and further reinforces the interaction. The combined actions of the two domains of Rtt106 enable it to perform the chaperoning function efficiently and properly. This is the first time anyone has described this mode of specific association between a histone chaperone and a modified histone complex.

Other authors of the study are Dan Su, M.D., Ph.D.; Qi Hu, Ph.D.; Qing Li, Ph.D.; James Thompson, Ph.D.; Gaofeng Cui, Ph.D.; Ahmed Fazly, Ph.D.; Brian Davies, Ph.D.; Maria Victoria Botuyan, Ph.D.; and Zhiguo Zhang, Ph.D.; all of Mayo Clinic. The study was a collaborative effort between the groups of Dr. Mer and Dr. Zhang and was funded by the National Institutes of Health and Mayo Clinic.

But when you're <a href="http://www.mtv.com/music/artist/bieber_justin/artist.jhtml">Justin Bieber</a>, you get shouts from Usher, Lil Wayne and Sean Kingston. Be jealous! On Thursday (March 1), the Biebs turned 18 years old, and his most famous friends hooked up with MTV News to celebrate <a href="/news/articles/1680191/justin-bieber-birthday-fan-questions.jhtml">Bieber's birthday</a>. "I hope that you are around all of your loved ones and having a good time. DENVER ? The Denver area averages 300 sunny days a year. If that has you worried about getting skin cancer, you might want to up your intake of vitamin A.

New research suggests people taking vitamin A supplements were 60 percent less likely to develop melanoma.

Denver dermatologist Dr. James DeVito said vitamin A?s benefits are well known.

?We use it because it has a polishing effect on the skin, it can smooth out the skin, it helps with fine lines and wrinkles, it also helps with acne,? DeVito said.

But for the anti-cancer effect it?s not topical vitamin A, it?s internal supplements that make the difference. So how much should you take?

?The recommended daily allowance for men and women is between 700 and 900 micrograms per day,? DeVito said.

You can, however, have too much of a good thing.

?Too much vitamin A can cause vision problems, it can also cause bone loss, osteoporosis,? said DeVito.

The study also said vitamin A in food didn?t contribute to the benefit, it was just supplements. DeVito suggested talking to your doctor about what daily amount is right for you.

March is Colorectal Cancer Awareness Month | Iloilo Bloggers

Public urged: Undergo early screening to prevent colorectal cancer

A Cebu-based cancer center urged the public to undergo early screening to fight colorectal cancer.

The Eduardo J. Aboitiz Cancer Center (EJACC) of the Ramon Aboitiz Foundation Inc. (RAFI) also advised the people to observe a healthy lifestyle through regular exercise and maintenance of a well-balanced diet.

EJACC urged the public to take this counsel seriously as the country celebrates Colorectal Awareness Month in March.

Colorectal cancer, a disease in which cells in the colon or rectum become abnormal and divide without control, forming a mass called a tumor, is one of the leading causes of cancer-related deaths among Cebuanos, based on the Cancer Registry of EJACC.

The Metro Cebu Population-based Cancer Registry of EJACC recorded 653 cases in 1998-2002 and 364 reported who have died of it. In the year 2002-2007 the Cancer Registry recorded an increased number of reported cases in Metro Cebu?862 cases were reported and 489 cases died of the disease.

Individuals age 40 and above, who have a sedentary lifestyle, who are eating high fat and low fiber diet, who have family and medical history of the disease, and who have polyps and ulcerative colitis, face high risk of acquiring colorectal cancer.

?The most important thing to know about colorectal cancer is that it often exists without any symptoms. This is why regular screening is very important. Regular screening can detect polyps at an early state before cancer develops or when it is most curable,? Ronald delos Reyes, EJACC program coordinator, said.

He also pointed out that the colon and rectum, which comprise the large intestine, are vital in turning liquid stool into formed fecal matter.

Colorectal cancer is characterized by a change in bowel movements, blood in the stools, abdominal discomforts, unexplained appetite and weight loss, fatigue, and pelvic pain in the later stage.

Diagnostic procedures in detecting colorectal cancer include fecal occult blood test, rectosigmoidoscopy, colonoscopy, irigography, virtual colonoscopy, fecal DNA test, and double contrast barium enema. The disease may be treated through surgery, chemotherapy, and radiation.

?The disease could be prevented by being health conscious and submitting to regular screenings,? Delos Reyes said.

ISPI?s Award of Excellence Program honors outstanding student and professional research. William L. Solomonson, CPT, PhD, of Wayne State University will receive an Award of Excellence for Outstanding Student Research for his dissertation which investigated the role of factors contributing to trust in the client-consultant relationship. He will receive the award at THE Performance Improvement Conference Saturday, April 21, 2012.

A core competency required for a performance consultant to be effective is the notion of ?partnering skill? and the ability to build trust with clients.? In this light, Solomonson explored the effect that perceived level of expertise, shared values, and the sharing of meaningful information have on trust in the client-consultant relationship. In the context of performance improvement, Solomonson determined much of the research effort has been placed on the ?technical? aspects of performance rather than the human relationship experience that occurs in the client-consultant interaction. Focusing on the ?relationship-side? of consultancy has particular relevance today with an increasing interest in performance consulting, as well as a lack of attention on the relational ?partnering skill? aspects of consultancy.

Improving Contributions of Consultants
A focus on trust is one means of potentially improving the contributions of consultants who focus on training, instructional design, or performance improvement. However, the client-consultant relationship is not commonly empirically studied; much of the extant business management client-consultant literature is conceptual in nature.? Additionally, the buyer-seller relationship of the client and consultant is generally thought of as being representative of a relationship between firms rather than between people; this does not translate well on the interpersonal level.? Through his research, Solomonson attempted to bridge these empirical gaps.

In his study, Solomonson quantitatively measured using a post-test only randomized experiment in which participants were randomly assigned into one of eight groups. Participants were individuals from selected undergraduate level classes at Wayne State University?s Department of Communication, and Oakland University in the School of Education during the fall semester of 2010 and winter semester of 2011.? Participants were randomly assigned to one of eight scenario-based questionnaires which measured the effect of three manipulated variables ? perceived level of expertise, shared values, and sharing of meaningful information ? on trust as a mediator to relationship commitment. The ability to trust, the comparison level of the alternative, and dependence were measured as control variables.

Influencing Trust
The findings of Solomonson?s research suggest that PI consultants can pro-actively manage the relationships with their clients by focusing on the key factors that influence trust and thus ultimately affect overall relationship commitment. For example, Solomonson concluded that if we know that a client?s perception of a consultant?s level of expertise has an effect on trust, then a consultant can act pro-actively to reinforce and communicate those aspects of expertise that reflect a high level of technical competence and knowledge to the client.

Solomonson?s dissertation research and was completed in August, 2011, at Wayne State University under the advisement of Committee members Dr. Ingrid-Guerra Lopez (Chair) and Dr. James L. Moseley.
